Do NAD+ boosters actually do anything?

NAD+ genuinely declines with age, and NR or NMN supplements reliably raise it back up within weeks in human trials. Six studies later, though, none has shown that a higher NAD+ reading translates into better blood sugar, body composition or a longer life.

02What's being claimed

Advocates argue that since NAD+ measurably declines with age and drives hundreds of metabolic reactions, restoring it with over-the-counter NR or NMN should reverse cellular ageing and restore a more youthful metabolism. The idea traces directly back to rodent longevity research and a Harvard scientist's own daily supplement routine.

The underlying chemistry is not in dispute. Rajman et al.'s 2018 review in Cell Metabolism confirmed NAD+ concentrations fall with age across species and tissues, and that restoring them with NAD+ precursors improves healthspan markers in animal models 1. Human trials back up half of that story: Conze et al. found NR supplementation doubled NAD+ metabolome markers in humans within eight weeks, safely and reproducibly 3.

That scientific premise met a famous champion. David Sinclair, whose Harvard lab had run the foundational rodent studies, publicised his own daily NMN regimen in his bestselling book and on the podcast circuit, turning a lab biomarker into a consumer routine. From 2022 onward, TikTok longevity content and a wave of direct-to-consumer NMN and NR brands pushed the trend further, treating a rising blood NAD+ number as proof of measurable anti-ageing progress, even though that proof was never tested in the trials that followed.

If you want to try NAD+ boosters, treat them as a biomarker experiment, not an anti-ageing protocol.

Raising NAD+ is real and safe at studied doses; a proven metabolic or longevity benefit is not.

01Treat NR or NMN as a low-risk supplement that raises a blood biomarker, not a validated anti-ageing treatment.
02Stick to studied dose ranges: up to 2,000mg/day for NR and up to 1,200mg/day for NMN, for periods of 12 weeks or less.
03Do not expect measurable changes in blood glucose, insulin sensitivity or weight; the largest trials found none.
04If you are over 60 and focused on blood pressure, note the modest diastolic effect seen in one meta-analysis, not a cure.
05Prioritise exercise, sleep and diet quality first, since these have the stronger evidence base for metabolic and healthspan benefit.
